Their 29-year-old sister had been stabbed in the arm.
The three are in Westmead Hospital where the younger brother is undergoing surgery.
The two others are stable.
A 57-year-old man was arrested at the home and taken to hospital under police guard.
He is being questioned by detectives at Blacktown Police Station and is expected to be charged with malicious wounding.
It is not clear how he is related to the siblings, but officers say he lives with them at the home.
Chief Inspector Bob Fitzgerald said a crime scene had been set up and forensic officers were combing the area for evidence.
